چکیده
Solid-state nuclear magnetic resonance (NMR) spectroscopy is an atomic-level method to determine the chemical structure, 3D structure and dynamics of solids semi-solids. This Primer summarizes basic principles NMR as applied wide range solid systems. The spin interactions effects fields radiofrequency pulses on spins in solid-state are same liquid-state spectroscopy. However, because orientation dependence state, majority high-resolution spectra measured under magic-angle spinning (MAS), which has profound types pulse sequences required extract structural dynamical information. We describe most common MAS experiments data analysis approaches for investigating biological macromolecules, organic materials inorganic solids. Continuing development sensitivity-enhancement approaches, including 1H-detected fast experiments, dynamic polarization ultra-high fields, described. highlight recent applications chemistry. ends with a discussion current limitations well areas points emerging this sophisticated used semi-solids biology, chemistry sciences.

A number of arylamides have been synthesized and found to exhibit potent antimicrobial activities against a broad spectrum of Gram-positive and Gram-negative bacteria while exhibiting low toxicity toward eukaryotic cells. These facially amphiphilic foldamers have a relatively rigid intramolecular hydrogen-bonded arylamide as a framework, which places trifluormethyl versus positively charged ami...
متن کاملAfterglow Solid-State NMR Spectroscopy.
Biomolecular solid-state NMR experiments have traditionally been collected through detection of 13C or 15N nuclei. Since these nuclei have relatively low sensitivity stemming from their smaller gyromagnetic ratios relative to 1H, the time required to collect multi-dimensional datasets serves as a limitation to resonance assignment and structure determination. One improvement in the field has be...

Solid-state Nuclear Magnetic Resonance can provide detailed insight into structural and dynamical aspects of complex biomolecules. With increasing molecular size, advanced approaches for spectral simplification and the detection of medium to long-range contacts become of critical relevance. We have analyzed the protonation pattern of a membrane-embedded ion channel that was obtained from bacter...
متن کاملStructural studies of proteins by paramagnetic solid-state NMR spectroscopy.
Paramagnetism-based nuclear pseudocontact shifts and spin relaxation enhancements contain a wealth of information in solid-state NMR spectra about electron-nucleus distances on the ∼20 Å length scale, far beyond that normally probed through measurements of nuclear dipolar couplings. Such data are especially vital in the context of structural studies of proteins and other biological molecules th...
متن کاملRNA structure determination by solid-state NMR spectroscopy
Knowledge of the RNA three-dimensional structure, either in isolation or as part of RNP complexes, is fundamental to understand the mechanism of numerous cellular processes. Because of its flexibility, RNA represents a challenge for crystallization, while the large size of cellular complexes brings solution-state NMR to its limits. Here, we demonstrate an alternative approach on the basis of so...
